In biochemistry agar and agarose gels have been used both as supports for electrophoresis [ 85 ], and for protein immobilization [ 86, 87 ], because of some favorable functional properties. In water, agarose is a typical strongly hydrophilic, lyophilic and extremely inert colloid.
Agar is isolated from the algae as an amorphous and translucent product sold as powder, flakes, or bricks. Although agar is insoluble in cold water, it absorbs as much as 20 times its own weight. It dissolves readily in boiling water; a dilute solution is still liquid at 42 °C (108 °F) but solidifies at 37 °C (99 °F) into a firm gel.

1% agar solution solidifies at 32 ~ 42 °C, its gel has elasticity, and its melting point is 80 ~ 96 °C. Many of its uses take advantage of this large difference between gelling and melting temperature. When compared with gelatin gels, it is tasteless, odorless, and sets more firmly, even at room temperature. Gelatin gels melt around 35-40 °C.

Agar is good for identifying and isolating healthy strains and cloning from tissue. Liquid culture is good, once you've isolated a good strain, for creating a large amount of inoculating medium for many grain jars or bags. They're a different growth medium for cultivating mycelium.
